泛素连接酶 384孔PCR芯片可以检测370个可以作为潜在药物靶点的泛素连接酶基因的表达情况。泛素化通过泛素蛋白酶系统调节细胞蛋白的降解过程。这个过程涉及到泛素激活酶(E1)、泛素连接酶(E2)和泛素连接酶(E3)的顺次作用。哺乳动物基因组编码大约12种E1酶,30-40种E2酶和600种E3酶。这些酶靶向许多蛋白底物,从而参与了许多生物学通路。此外,泛素化并不总是靶向降解的蛋白。例如,细胞表面的跨膜受体的单泛素化可以作为细胞内吞作用的标记,而多泛素化则作为泛素结合蛋白的支架。泛素蛋白酶降解通路的混乱与多种疾病相关,例如癌症、糖尿病、心血管疾病和神经退行性疾病。E3酶的多样性和底物特异性使它们在正常的以及疾病相关的通路中发挥着重要的作用,也使它们成为了潜在的药物靶点。进行药物筛选分析,不仅需要检测药物靶点的表达,还需要检测有冗余或补偿功能的基因的表达状况,因为这些基因会降低药物的功效或者是引起脱靶反应。这款芯片涵盖了来所有重要的的E3家族泛素连接酶,从而可以聚焦于那些研究数据充分,表达广泛的酶。通过实时定量PCR的方法,研究者即能够利用该芯片简单可靠地同时检测泛素连接酶相关基因的表达。
