小鼠模型是结肠炎造模常见模型,自从1985年首次采用葡聚糖硫酸钠(dextran sulphate sodium, DSS)制备出仓鼠溃疡性结肠炎模型后,已有大量数据证明DSS结肠炎模型的病因、临床症状、病理改变及治疗应答均与人类溃疡性结肠炎(ulcerative colitis,UC)相似。
现行研究中常用的UC建模类型大约分为:自发模型、诱导模型(化学药物诱导:TNBS三硝基苯磺酸,DSS葡聚糖硫酸钠等、免疫学方法诱导、细胞移植诱导)以及基因修饰模型等。
DSS造模优势
葡聚糖硫酸钠盐(Dextran Sulfate Sodium Salt, DSS)是一种聚阴离子衍生物,其诱导结肠炎模型的机制虽仍未十分明确,但通常认为与巨噬细胞功能失调、肠道菌群失调、DSS对结肠上皮的毒性作用、细胞因子在DSS结肠炎模型的发病中起重要作用等机制有关。
对比多种UC建模类型,DSS造模具有很大优势:
1、症状与人类UC高度相似:可用于研究结肠炎发生机制和药效研究;2、成模率高:自由饮用DSS水溶液,简单易行、重复性强;3、可构建多种结肠炎模型:急性结肠炎、慢性结肠炎,还可以联合AOM构建结肠炎相关性癌症(CAC)模型;4、适用于多种属动物造模:小鼠、大鼠、斑马鱼、猪、果蝇等;5、安全性高:DSS可被自然生态系统降解,对环境安全。

DSS建模实验步骤
图1. 小鼠结肠炎造模方案 DSS浓度:急性期模型通常为3-5%,慢性期模型通常为1-3%;可根据实验需求及动物类型摸索浓度。该浓度为质量体积比,使用饮用水配置后,0.22µM滤膜过滤除菌。
数据记录:每隔24h记录小鼠体重、粪便粘稠度、粪便潜血等指标。

急性DSS结肠炎模型
Day1:称重并标记各组C57BL/6小鼠。待造模小鼠饮用3-5% DSS水溶液,未处理组小鼠饮用正常水。Day3:给予待造模小鼠更换新鲜3-5%DSS饮用水。Day5:给予待造模小鼠更换新鲜3-5%DSS饮用水。Day8:给予待造模小鼠更换新鲜不含DSS的饮用水。

慢性DSS结肠炎模型
Day1:称重并标记各组C57BL/6小鼠。待造模小鼠饮用1-3% DSS水溶液,未处理组小鼠饮用正常水。Day3:给予待造模小鼠更换新鲜1-3%DSS饮用水。Day5:给予待造模小鼠更换新鲜1-3%DSS饮用水。Day8:给予待造模小鼠更换新鲜不含DSS的饮用水。Day22-Day26:重复Day1-Day5操作。Day29:给予待造模小鼠更换新鲜不含DSS的饮用水。Day43-Day47:重复Day1-Day5操作。Day50:给予待造模小鼠更换新鲜不含DSS的饮用水。
